New players on PCa- DCVax (vaccine) in trials vs. Provenge & Jevtana/Cabazitaxel (FDA-ok'd)

New vaccine is in trials and is little different but similar with dendretic cell action used, may compete well with Provenge. The active ingredient in both companies' vaccines is dendritic cells. That is an active pharmaceutical ingredient, it's the active agent that's doing the job. But according to data provided by Northwest Biotherapeutics, the percentage composition of Dendreon's product by the company's own published material is 15% antigen presenting cells -- dendritic cells and others. According to NWBO's published material, DCVax(R) is over 80% active ingredient. The official product release criteria are over 80% and Northwest Biotherapeutics' platform usually hits over 90%, thus the active ingredient in NWBO's vaccine is much more concentrated.

Third factor: How is the product administered to the patient

According to Powers, because there is such a low percentage of active ingredient in Dendreon's vaccine, they have to deliver a huge volume, and the only way to do it is intravenous infusion, which is how they deliver Provenge to their patients.

This IV delivery can take over an hour. By contrast, because the active ingredient is so concentrated in the DCVax(R) vaccine, that NWBO only needs to administer a tiny volume of "just a few drops" and it's administered with a simple injection under the skin, like a flu shot or an insulin shot.

Now on Cabazitaxel suggest you search and read up on it as a new chemo therapy drug for PCa and is used likely in conjunction with a steriod. Jevtana is the name also on this.

Trial phase III- 750 men whom failed both hormone therapies and chemo with taxotere and divided into 2 groups: 1st Cabazitaxel with Prednisone (a symptoms relief thing I believe); 2nd group Mitoxantrone with Prednisone. Treatments every 3 weeks for 8 cycles studied for overall survival and seondly outcomes including psa changes. Results: overall suvival with Cabazitaxel improved by 2.4 months (from 12.4 to 15.1), however Caba. group tolerated 6 cycles and the Mitoxantrone(naovantrone group) tolerated 4 cycles.

Toxicity is different and worse for Caba. especially grade 3/4 neutropenia (81%) with increased febrile episodes is very relevant. (looks like a trade off in that area)

I have not looked into either of these much, just mentioning them for others, do your own searches and read up on them. Someone else can add links and discussion to any of this and probably should.

Also the newer FDA approved LHRH drug is likely available easily now, Degarelix. Unlike Lupron, Zoladex, Precisis, Eligard, Trelstar and other common LHRH, this one has no PCa flare issues and castrate levels in 24 hrs., instead of days, and casodex is not a necessary pre-drug to use to avoid possible flare. I hope someone herein can elaborate on it and possible side effects for them, maybe cost if known too.
