Has anyone heard of or know of anyone using drug(s) to target the mTOR pathway? For PC or any other type of cancer...

It's my understanding that this pathway is central to cell growth and survival.

What do you want to know about mTOR inhibitors -- are you on a clinical trial of one? In a recent RCT, everolimus+casodex didn't provide incremental benefit. Probably it's necessary to block several pathways simultaneously with a cocktail of inhibitors. Combination with Depakote is especially intriguing because they're both already approved for other uses.

Tall Allen,

No clinical trial, but have been prescribed a low dose of Rapamycin, Resveratrol, and Metformin. Also known to be a possible anti-aging cocktail.

It's a powerful immunosuppressant, but as long as your doc says it's OK and is monitoring you. Do you think it is keeping your PSA low?

This doctor is an oncologist? As I thought about it, you mentioned "low dose" rapamycin. With immune system modulators, dose can have paradoxical effects. For example, cyclophosphamide is used in normal doses to suppress the immune system, and is sometimes helpful for autoimmune conditions like osteoarthritis. However, in low doses, it activates the immune system and is used in cancer therapy. Tricky stuff.

My MO is Dr. Myers. He monitors everything monthly. Is it keeping psa low? Hard to say. Last Lupron was about a year ago, avodart as maintenace, Testosterone back to normal for the past 6 months. All other markers and scans look ok

What dose are you taking? Any observed increase in infections - colds, seborrhea, skin lesions? Does he monitor your white blood cells? I have a friend who has talked to Scholz about it - he was curious, but he's not the experimenter that Myers is, imho.

I'd be curious to hear Myers' opinion on Depakote (a powerful HDAC inhibitor). It's also a powerful mood stabilizer.

1mg daily....he increased resveratrol while using rapamycin...I read somewhere that it increases its effectiveness.

No colds, illness...feel great...minor rash on chest occasionally...

He makes me get blood work monthly which includes wbc. Mine hasn't changed since taking this medicine 4 months ago. My wbc has always been low...hovers around 4.

Just that you knew about some of the side effects is impressive...

I see him next month and will ask about depakote.
