Amitriptyline is generally prescribed as a mild antidepressant, so I'm not surprised it didn't help with your weight loss. Weight loss isn't even an off-label use or a side effect. Sometimes doctors will prescribe anti-depressants because they think we gain weight due to depression. This is usually incorrect. I know it can take two to three weeks for the medication to take effect, so would assume it would take about the same amount of time to clear your system.
Please do let us know what your thyroid panel shows, including the reference numbers. It's really difficult to identify thyroid issues solely from symptoms, because the symptoms of thyroid problems are pretty non-specific.
