The UNITI-2 psoriasis Stelara dosages by injection at 80 mg were ineffective for Crohns, 4 times a year is not enough. They kept experimenting with different dosages and different frequencies.
In the UNITI-2 study they cranked it up to 400 mg by infusion was when things started happening
Back in 2006 First 80 mg did nothing, Second 80 mg a week later nothing, Third 80 mg was the one that started turning things around. Note that this now was a cumulative 320 mg in three weeks. By the Eight injection it blasted Crohns good, it was utterly amazing. By, then it was a cumulative 640 mg.
Realize that in 2006 Stelara dosages were unknown to what will happen with humans, I was the 26th person in the world to be tested with this. That is why they kept cranking little by little, week by week, a little more instead of a big hammer 400 mg infusion. I had blood and urine tests every week, the liver was one of their focus points. If things would have gone south they would have stopped the injections immediately.
That is why if they get you on Stelara make sure that is more than a 80 mg injection. If it is at 80 mg and nothing changes it is not Stelara's fault. Stelara like the others start with a loading doses, usually 3 doses. If you see some results great, but the maintenance doses should be more than 80 mg every other month.
With Entyvio had my 3 loading doses, nothing. Another maintenance dose 2 months later, nothing, Another two months later, nothing (up to 5 by now). I keep getting told that Entyvio works very slow, so by the 6th dose I was ready to talk with the Stelara researcher which I had to wait 2 months to see him.
By then I had my 7th dose, nothing. The moral of this story: these drugs may be slow acting but you should see SOME CHANGE by the 5th dose, if no changes by the 6th and certainly the 7th there is a high probability that it is not going to work.
