Azlocillin- 99.9% eradication like Disulfiram

Azlocillin sodium is another drug to watch as just like Disulfiram it completely sterilized Borrelia in vitro in that Stanford study. Currently it's only available overseas and in IV form but a company called Flightpath biosciences is working on an oral formulation and says it has demonstrated in vivo efficacy in mice. This could be a good alternative to Disulfiram as it doesn't carry some of the risks associated with DSF and doesn't require a whole lifestyle change. Stay tuned!

Yes really interesting. I saw the interview with Dr Rajadas talking about it. I had never heard of it but if I have any trouble or treatment failure with Disulfiram, I am going to look into next. Probably very cheap in India and some East Asian countries.

It's only available via IV overseas but Hopkinton drug turned Rocephin into an oral form using liposomes so I called them and they are interested but are having trouble finding it in formularies. If anyone had any idea on how they can get there hands on it you can call them or let me know and I will. The more interest they see the more they will be motivated to delve into this further. Also gotta see if it will cost effective to make but I'm pretty sure they will have the capability of making it in liposomal form.

Yeah I’m very interested in it as well, hopefully they find some novel drugs for Babesia and Bartonella as well, since most of us have all trifecta.

If so, it was from 2016 - wouldn't it have been more regarded in recent time if it were *that* effective? Not mention it seems hard to acquire... I dunno i'm just thinking aloud here

DeadMan- That is the study and also is the one with Disulfiram that is just now gaining traction. It takes time to develop oral versions of drugs that are only in IV form and then there's the time it takes to conduct studies on mice or rats considering that study was all in vitro.

JovaLyme- You bring up some good points. We do need more effective drugs for co-infections. The prevailing thought with this disease is that once you take care of Borrelia your immune system can take care of the rest. Not sure if anyone knows how true that is but I have two friends that were cured with Rocephin and one with Doxy that caught theirs early. If it weren't true then that means none of them had co-infections which I highly doubt so I think there's some truth to that.

Early cases have a much higher success rate - and yes just lyme treatment often works.
But for chronic Lyme - many (most?) docs generally believe it’s the coinfections that need to fall.
That is what DrJ in DC says - especially Babesia - he goes after it first (after a stabilization period)

I know no one with chronic Lyme that treated just lyme and then their immune system took care of bart and Babs.

I would run the other way if a LLMD told me he wouid treat lyme and then my immune system would be able to take care of bart and Babs.

Well if someone can explain why early treatment with Rocephin and even Doxy is successful with co-infections present I'm all ears. I've even seen it with chronic Lyme mostly with Rocephin and a few cases with Vanco. The current oral abx are just not effective at eradicating stationary phase Borrelia and persister round bodies. Disulfiram is showing promise but a bit risky with all the neuropathy I'm seeing. Better to wait on the new formulation IMO.

Because it’s EARLY - that’s why.
If you treat early - even with orals - the infection(s) haven’t had time to flourish and get into the brain and nerves...
The bacterial load is low

I’ve not heard of cases where chronic Lyme was treated and Babs fell.
Bart - maybe - because some of the lyme abx cover bart too.

Can confirm! this is why you can usually crush borrelia like nothing with doxy when you hit it right after it's been inoculated in the body (and before it has truly disseminated).

But once it's in the CNS, hoo boy, what an effort to push it back. Not impossible, but yes, an indescribable effort for some of us, especially those of us who are prone the strong immune system responses that occur with die-off...

I agree with all of you, we need drugs that penetrate the CNS/BBB. that eradicate each of the three B’s and preferably bacteriosidal not bacteriostatic. It’ll take at least one drug for each is my guess, we’d have to be very fortunate to get a 2 for one even if Disulfiram seems promising for both Borrelia and Babesia. As was mentioned above it’s maddening that most of the studies are only on Borrelia when almost everyone that stays sick has at least 2 infections and many have way more.

There is a set of answers out there and over the passed few years major strides have been made in research, we just need to keep going in the same direction with tests for each of the three B’s, antibiotics that cure each of them. It’s all about giving research money to people that produce results like Ying Zhang, the group at standford that tested disulfiram and this drug, Eva sappi and Alan McDonald I’d he’s still around. Hopefully there’s new up and coming scientists that become interested in TBD/VBD because of the scale of the problem or because it’s effected them personally.

Imo there is a lot to be excited about, disulfiram, azlocillin, likely more promising agents to come (even things like gallium nitrate and methylene blue for bartonella)... patients with late stage disseminated lyme are not dead in the water. We just have a larger hill to climb. And there is more research being done evaluating agents for babesia, so we are likely going to be getting better knowledge on what actually works and doesn't work. Mamoun Ben Choukri's babesia team is currently conducting a study evaluating disulfiram and other agents in vitro and in vivo activity on babesia strains microti and duncani. So more info is right around the bend, at least regarding babesia!!! (and we know current mainline therapeutics for babesia leave a lot to be desired, corroborated by Choukri's study last year).

Disulfiram seems like a great two-fer regarding borrelia and babesia. That said, for patients with heavy CNS involvement will likely require the golden rule approach, ie treat to tolerance (with great care to not stir up overwhelming flare responses).

Like Chiledog said, we have to try to keep supporting those teams that are really looking to help lyme patients, the teams that are looking for answers. Zhang's team, Sapi's team, Rajadas's team, Kim Lewis's team... just those 4 teams have provided a wealth of knowledge for lyme patients, pretty extraordinary in fact.

I agree. I had been stockpiling Disulfiram from my doctor and online sources and, in the back of my mind, I was cautiously optimistic that I could be finally cured. I was dealt a blow with neuropathy only a week into treatment and now I don’t want this cold foot to get any worse...very upsetting.

Last few days, I have been researching Azlocillin and there seems to be no easy pathway to treatment...although a possibility I am hoping. This post has cheered me up a bit though and I guess I have waited for a decade so what is one more year of low function and constantly making excuses to those around me about my lack of energy and dull personality. The new Disulfiram-like compound could be a much better option...if it is realized.

Azlocillin works better with silver nanoparticles.
Which is interesting if you compare it to Disulfiram and copper.
I have a contact that can, is willing to (and has) synthesized Azlocillin sodium.
I'm willing to try it, however, I don't know if it would work intramuscularly.
As I don't want to do my own IV.
When I read about IV abx, it always makes me wonder why intramuscular antibiotics are not done more often.

These can easily be done in the leg by patients themselves.
Just like any other at-home injection.

IV abx go directly into the bloodstream resulting in instant bioavailability.

IM - it’s held in the muscle and slowly releases it into the bloodstream

I have been reading a lot. It seems there are sources of Azlocillin that would cost about $1,000 daily...if I can get them to ship to me. I cannot afford it.

I heard Dr Rajadas mention Azlocillin but that is all I have heard so far. I have not heard the compound that came in third overall for their team.

Doxorubicin sounds risky.

Josamycin could be worth a try but no mention of it for Lyme anywhere...that I have seen.

Yes I certainly would! I injected Benzylpenicillin for a few years without major issue. I was truly devastated to get neuropathy from Disulfiram and I am looking for hope. From what I can see, it is cost prohibitive but I am praying that will change somehow. Eleven years of this is long enough...wearing me down. All of my LLMDs over the years have given me credit for continuing to work full time and know I am not a victim kind of guy but please universe throw me a freakin bone! Soon☹️
