The research on genetic susceptibility to UC has focused only on nuclear DNA. Some new research has found that mitochondrial DNA also plays a role. I think this is bad news because we will soon have gene treatments for nuclear DNA. I don't think there is much active work on editing mitochondrial DNA.

Is mitochondrial DNA connected to IBD? In my case, although no one ever talked about it, my father's relatives seem the likely source for my UC, if there is a genetic proclivity. However my mothers side has the Irish genes of higher incidence. No one in my family ever declared having IBD but pretty sure it's the English genes based on lifestyle of grandmother and great-grandfather.

My 23andme gene results (done before FDA stopped it) say Crohn's is higher chance but my diagnosis is UC.
Also very high chance of celiac but office test said no and never had the internal celiac test. Removing gluten has been very important to my symptoms.

When we collected the first 5,000 DNA samples from participants of the 23andMe IBD community, we took an early look at the data to see how it compared to the data of other high-quality, more conventional (not online) IBD genetic studies. We identified genetic variants (differences in DNA) that are likely to either increase or decrease the risk of developing IBD.

We were excited to see that all 26 genetic variants discovered through this analysis replicated previous signals from the more conventional studies.

This gives us confidence that the 23andMe model of research (all conducted online and at home) will soon give us new insights into the genetic basis of IBD.

IBD has many links to nuclear DNA, known for decades.

This research ALSO finds links to mitochondrial DNA. So it is BOTH. Having some mitochondrial DNA association in some cases does not invalidate or change the history you are seeing (for nuclear DNA) on your father's side of the family.

I think 23andMe only looked at nuclear DNA.

Not sure its a big issue. The genes in the mitochondria wont play nearly as big a role in IBD pathogenesis as the nuclear DNA.

I'm just glad they're exploring all possibilities on the table. Better to have a large set of tools to solve this, rather than just several tools. It'd increase the odds of a eureka moment.

1) The sheer number of genes total and associated with IBD in each 2) Mitochondria dysfunction hasnt really been implicated in IBD before and most mito genes are involved in some way with mito function

I agree there are more non-Mitochondria genes. But Mitochondria genes have now been implicated in IBD.

On the number of genes basis, the problem concern may be small. On the fact that we can edit nuclear DNA and I am not sure we can edit Mitochondria DNA, I said it might be bad news.

When I get a chance I will look at exactly what Mitochondria genes they found.
