The initial clinical studies for oral mesalamine included it being taken multiple times a day. The theory for multiple-times-per-day is that the pills will arrive at different times in the large intestine and release their medication over a wider range of hours for optimum healing affect. I have some qualms with that theory as the digestive tract is full of valves and holding areas. Even though you might take pills at 8:00am and 8:00pm, there's no guarantee they arrive at much different times in the large intestine (might even arrive at the same time).
Lialda/apriso were studied with once-per-day administration, and it is their claim to fame. Proved to be just as effective with their delayed-release formulation.
Taking pills multiple times per day can be a pain to remember and can lead to missed doses. I'd say do what works best for you, if you know you are going to miss doses with multiple application then just take them when you're going to remember (all at once, evening and night, etc.). I'd say taking all of one's pills each and every day is more important than the split doses are.
